Protein Characterization

Beside proteomics Proteome Factory offers since many years contract research for complete sequencing of proteins like antibodies and recombinant or therapeutic proteins by a comprehensive protein sequencing approach which includes N-terminal and internal Edman protein sequencing, capillary LC separation of proteolytical generated peptides and mass spectrometry including de novo peptide sequencing.
In additon Proteome Factory offers characterization of proteins and antibodies by most modern mass spectrometric techniques in combinantion with HPLC and N-terminal peptide sequencing (Edman degradation). Proteins are digested by different endoproteases and/or chemical cleaving methods to generate overlapping peptides of the protein. Following HPLC separation of the different digests the peptides are sequenced by Edman degradation, analyzed by ESI-MS and MS/MS techniques. To ensure highest sensitivity and reliablilty Proteome Factory uses nanoLC-ESI-MSMS which allow de novo peptide sequencing by MS/MS analysis in combinantion with de novo sequencing software and other bioinformatic tools. By combinantion of the peptide sequence data the protein's sequence is determined.
